Last week I brought a vacuum pump to bleed my brakes without realising I needed air pressure unit for it to work, I decided to use it anyway with the manual- open nipple, pull lever, close nipple, release lever method, and it worked very successfully.

However my question is, will this method work with the CBS fluid? Will pulling the the rear lever push the fluid out of the nipple the same as the front brakes?

Yeah, you can pull/push fluid out that lower nipple, same as the front. It has a separate reservoir and MC from the front(right lever), though.
